Hi,
I need to create a database connection for my cloud acumatica ERP instance. Is it possible or how can I do it?

What do you mean by DB connection? Do you mean Accessing the Cloud SQL Directly outside Acumatica like if you would have it on your Local Server? If yes, then not possible. You can only communicate with Cloud applications including Acumatica using API. Otherwise please clarify.
