Transaction speeds for large SKU base

Has anyone experienced decreased overall operational speed with a large data set of >500 SKUs (i.e., is there inverse relationship between operational speed and SKU quantity in a system)?

We have several clients with 5000+ sku for inventory. With no noticeable degradation in performance.

The only exception was one client had large Sales Orders with more than 500 lines. This seems to be more of an issue than the number of Sku’s in the database. Somewhere we heard about a method to limit how many lines load at a time on the Details tab but we never implemented it. The client decided to split these large orders instead and group bill the shipments on the invoice later.
